Selank: The Russian Nootropic Peptide for Anxiety and Cognitive Enhancement

Selank is a synthetic heptapeptide analogue of the endogenous peptide tuftsin (Thr-Lys-Pro-Arg). It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and has been used clinically in Russia and Ukraine since 2009, approved there as an anxiolytic and nootropic under the brand name Selank.

Mechanism of Action

Selank works through multiple pathways that together produce anxiolytic effects without sedation:

  • BDNF upregulation — increases Brain-Derived Neurotrophic Factor, promoting neuroplasticity and new synapse formation. This is the likely mechanism behind cognitive enhancement.
  • GABA modulation — enhances GABAergic tone (similar mechanism to benzodiazepines but without receptor binding or dependence risk)
  • Serotonin metabolism — modulates serotonin without direct receptor agonism, reducing anxiety through a different pathway than SSRIs
  • Enkephalin stabilisation — inhibits the enzymes that break down enkephalins (endogenous opioids), extending their natural anxiolytic effect
  • IL-6 regulation — modulates immune inflammatory signalling, relevant for anxiety that has inflammatory components

What Makes Selank Different from Benzodiazepines

Benzodiazepines directly bind GABA-A receptors, creating strong sedation, cognitive impairment, and dependence. Selank modulates GABA tone without direct receptor binding — the anxiolytic effect is present without the sedative, cognitive-blunting, or dependency profile. Research subjects report reduced anxiety with simultaneously improved mental clarity and cognitive performance.
